as far as we experienced, even if supported formats for publishing in Amazon Book Store includes also epub and html (formats ), we suggest you to create a KF8 (mobi) file, that address all the limitations imposed on Kindle devices. For example all interactions are removed to obtain a compliant mobi file.
Per associare più di una azione a un pulsante occorre selezionare il pulsante, scegliere l'evento e poi aggiungere le azioni clickando su Add new Action. In alternativa si può definire un oggetto "Action list" e poi associare all'oggetto bottone la Action List appena definita.
Nel progetto di esempio incluso in PubCoder ci sono esempi di entrambe le soluzioni.
Detto questo, il problema specifico invece mi sembra più complesso se affrontato in questo modo, perché ogni pagina non ha memoria di ciò che è stato eseguito in precedenza. Occorre inserire del codice javascript ad hoc.

Lo standard HTML prevede un oggetto chiamato "localStorage" che può salvare informazioni di questo tipo. Il suo supporto però non è garantito dagli EPUB reader.
A livello di app, sebbene non ufficialmente supportato, dovrebbe funzionare.

Il problema è che purtroppo non esiste al momento un EPUB reader per Android equivalente ad iBooks di iOS.
Il mio consiglio è di ovviare a questa mancanza esportando una app per Android e quindi pubblicarla sul Google Play Store
si potrebbero impostare le dimensioni dell'iframe in questo modo:

Questo ovviamente funziona nel contesto delle app (Android o iOS), mentre per epub occorre tener presente che molti reader bloccano il caricamento di qualsiasi risorsa esterna.
